چکیده مقاله:

Potato virus Y (PVY) is one of the most damaging viruses of potato plants which infecting most cultivars and causing significant yield and economical losses. The application of the concept of pathogen derived resistance opened new horizons for the development of virus-resistant plants. This research was carried out to study RNA silencing to engineered potato plants that are resistant to potato virus Y (PVY). RNAi construct was designed from coat protein (cp) segment of virus RNA. Potato explants were transformed by Agrobacterium tumefaciens AGL1 with pCAMRNAiCP containing bar gene. The rate of regenerated plants and gene transformation were evaluated. Results showed that 42 percent of regenerated plants were transformed. The expression of CP hairpin construct was determined by RT-PCR analysis. PVY resistance bioassay showed that 82 percent of transgenic potato were resistance to PVY.

Heterologous Expression of Potato Virus Y Coat Protein, Isolate Pot187

Background: The advent of recombinant DNA technology has facilitated heterologous expression of proteins from various sources in different host systems including Escherichia coli. If a plant virus coat protein is expressed in the bacterium it can be used as the antigen for antibody preparation. Such a recombinant antigen preparation can be particularly useful where equipment such as ultracentri...

Transient expression of coding and non-coding regions of PVY confer resistance to virus infection

One of the most efficient mechanisms by which plants protect themselves from invading virusesis the specific RNA-dependent silencing pathway termed post-transcriptional gene silencing(PTGS). In this mechanism, resistance to a virus is engineered through the expression of asegment of the virus genomein transgenic plants. Potato VirusY (PVY) is one of the mostdamaging viruses of potato, infecting...

Cloning and sequencing of potato virus Y coat protein gene from an Indian isolate and development of transgenic tobacco for PVY resistance

Potato virus Y (PVY) coat protein gene has been cloned from an Indian isolate of PVY. The coat protein gene from this isolate showed 93.2% nucleic acid sequence homology and more than 94% amino acid sequence homology compared to an American isolate. The coat protein gene has been sub-cloned into plant expression vector pBINPLUS and tobacco leaf discs were transformed using Agrobacterium tumefac...

Induction of resistance to potato virus Y strain NTN in potato plants through RNAi

Potato viruses cause enormous economic loss in agriculture production. Potatoes can be infected by a number of different viruses that affect negatively the harvest and the tuber quality. Direct and effective drugs against plant virus diseases are still not available and control is only applied as agricultural measures and pesticides against virus vectors. Potato virus Y (PVY) is transmitted by ...
